Lymph‐circulating tumor cells show distinct properties to blood‐circulating tumor cells and are efficient metastatic precursors
Study of MTLn3 and expression in lymph and blood ctc's (!) - use in vitro culture to expand before analysis I think. Also do some stemness/spheroid studies though not by facs. The leading cause of breast cancer‐associated death is metastasis. In 80% of solid tumors, metastasis via the lymphatic system precedes metastasis via the vascular system. However, the molecular properties of tumor cells as they exit the primary tumor into the afferent lymphatics en route to the sentinel lymph nodes (SLNs) are not yet known. Here, we developed an innovative technique that enables the collection of lymph and lymph‐circulating tumor cells (LCTCs) en route to the SLN in an immunocompetent animal model of breast cancer metastasis. We found that the gene and protein expression profiles of LCTCs and blood‐circulating tumor cells (BCTCs) as they exit the primary tumor are similar, but distinct from those of primary tumors and lymph node metastases (LNMs). LCTCs, but not BCTCs, exist in clusters, display a hybrid epithelial/mesenchymal phenotype and cancer stem cell‐like properties, and are efficient metastatic precursors. These results demonstrate that tumor cells that metastasize through the lymphatic system are different from those spread by blood circulation. Understanding the relative contribution of these cells to overall peripheral blood‐circulating tumor cells is important for cancer therapy. Whether these two types of cell occur in cancer patients remains to be determined.

Interplay between chemotaxis and contact inhibition of locomotion determines exploratory cell migration : Nature Communications : Nature Publishing Group
Used MTLn3-ErbB1 cells in microfluidic narrow channels with EGF gradients - I think the cells perturb the gradients. They have contact inhibited locomotion which cells touch each other. KD of EphB3 reduces this CIL. Can image rac activation, PH domain shifts, and lifeactin in the channels. "Our results suggest that both cues act on MTLn3 cells by impinging on a PI3K, Rac and RhoA signalling network, affecting actin polymerization and protrusion formation (Fig. 7). CIL cues are mediated in part by EphB signalling through engagement of ephrin-b ligands present on the surface of other MTLn3 cells.... Contrary to previous studies conducted within other cell types, where CIL was induced through RhoA, we suggest that, in MTLn3 cells, EphB signalling induces CIL through inhibition of EGF-mediated protrusion formation via suppression of PI3K, as the direct addition of clustered ephrin ligands did not activate RhoA and did not induce cell retraction.

Dormant but migratory tumour cells in desmoplastic stroma of invasive ductal carcinomas
data that MTLn3 cells in high density collagen are migratory but not dividing/dormant. Also does LCM on solitary (maybe tumor) cells in invasive ductal carcinoma of patients and finds upregulation of a number of cytoskeletal proteins and had upregulated antiproliferative genes.

Oncogene - p140Cap suppresses the invasive properties of highly metastatic MTLn3-EGFR cells via impaired cortactin phosphorylation
MTLn3 EGFR cells and effect of p140CAP. Unfortunately there are growth effects which raise concerns about the mechanism. There is a 3D spheroid assay but the methods are inconsistent it seems to me on how it was performed.

fes is a cytoplasmic tyrosine kinase, and its loss reduces the ability of macrophages to enhance MTLn3 3D invasion. Probably working mainly in the primary tumor to enhance angiogenesis and invasion.

Coronin 2A regulates a subset of focal-adhesion-turnover events through the cofilin pathway -- Marshall et al. 122 (17): 3061 -- Journal of Cell Science
MTLN3 study! nice coronin, cofilin movies. Coronin 2A binds to FAs and localizes slingshot/active cofilin to them which contributes to their turnover. Suppressing coronin causes larger and more stable FAs.
